Identification of Poinsettia Cultivars Using RAPD Markers

    Get PDF
Randomly amplified polymorphic DNA (RAPD) techniques were used to compare the DNA from leaf tissues of nine commercial poinsettia (Euphorbia pulcherrima Wild ex Klotzsch) cultivars. Amplification occurred in 57 out of 60 (95%) tested primers. Nine primers that revealed polymorphisms among cultivars were selected for further evaluation. Forty-eight RAPD bands were scored from these primers, and 33 (69%) were polymorphic. All tested cultivars could be discriminated with seven bands generated from primers OPB7 and OPC13. Results of a UPGMA cluster analysis and principal components analysis placed the nine cultivars into two groups: one group consisted of `Jingle Bells\u27, `Supjibi\u27, and `V-17 Angelika\u27, the other of `V-14 Glory\u27, `Red Sails\u27, `Jolly Red\u27, and `Freedom\u27. `Lilo Red\u27 and `Pink Peppermint\u27 belonged to the latter group, but were relatively distant from other cultivars in that group. These results indicate that RAPDs are efficient for identification of poinsettia cultivars and for determination of the genetic relationships among cultivars

The molecule of the title compound, C31H29N3, contains a hexyl chain, a coordination unit (benzimidazole) and a functional group (carbazole). The benzimidazole ring is not coplanar with either the phenyl ring or the carbazole system, making dihedral angles of 43.26 (3) and 39.03 (2)°, respectively. The dihedral angle between the phenyl ring and the carbazole system is 24.42 (3)°. The hexyl Cβ atom (with respect to benzimidazole) deviates by 1.124 (2) Å from the benzimidazole plane, although the Cα atom lies in the plane. The hexyl Cβ atom (with respect to carbazole) deviates by 1.315 (1) Å from the carbazole plane, although the Cα atom lies in the plane. The crystal structure is stabilized by intermolecular C—H⋯π interactions

Generation of Oligodendrocyte Progenitor Cells From Mouse Bone Marrow Cells.

Oligodendrocyte progenitor cells (OPCs) are a subtype of glial cells responsible for myelin regeneration. Oligodendrocytes (OLGs) originate from OPCs and are the myelinating cells in the central nervous system (CNS). OLGs play an important role in the context of lesions in which myelin loss occurs. Even though many protocols for isolating OPCs have been published, their cellular yield remains a limit for clinical application. The protocol proposed here is novel and has practical value; in fact, OPCs can be generated from a source of autologous cells without gene manipulation. Our method represents a rapid, and high-efficiency differentiation protocol for generating mouse OLGs from bone marrow-derived cells using growth-factor defined media. With this protocol, it is possible to obtain mature OLGs in 7-8 weeks. Within 2-3 weeks from bone marrow (BM) isolation, after neurospheres formed, the cells differentiate into Nestin+ Sox2+ neural stem cells (NSCs), around 30 days. OPCs specific markers start to be expressed around day 38, followed by RIP+O4+ around day 42. CNPase+ mature OLGs are finally obtained around 7-8 weeks. Further, bone marrow-derived OPCs exhibited therapeutic effect in shiverer (Shi) mice, promoting myelin regeneration and reducing the tremor. Here, we propose a method by which OLGs can be generated starting from BM cells and have similar abilities to subventricular zone (SVZ)-derived cells. This protocol significantly decreases the timing and costs of the OLGs differentiation within 2 months of culture

AbstractObjectiveTo study the change of serum C-reactive protein (CRP) levels and its correlation with ketosis in type 2 diabetes mellitus patients with acute ketosis intoxication.MethodsA retrospective analysis was conducted for the patients with type 2 diabetes mellitus from August 2015 to January 2016. The patients combined with ketosis were included into diabetic ketosis group and the patients without ketosis were included into negative control group. The clinical data were collected from two groups including general data, blood pressure, liver function and the levels of blood fat, glycosylated hemoglobin, blood ketone, β-hydroxybutyric acid and CRP. The discrepancy of clinical data between two groups was analyzed.ResultsThe levels of glycosylated hemoglobin [(11.6 ± 2.1)% vs. (8.3 ± 1.9)%], blood ketone [0.65 (0.3, 1.75) vs. 0.1 (0.1, 0.2) mmol/L], β-hydroxybutyric acid [0.595 (0.303, 1.775) vs. 0.08 (0.06, 0.15) mmol/L] and CRP [0.595 (0.303, 1.775) vs. 0.08 (0.06, 0.15) mmol/L] were significant higher than those of negative control group, while the levels of blood pressure, blood fat and aminopherase had no significant difference. The serum CRP levels showed positive correlation with blood ketone and β-hydroxybutyric acid (r = 0.490 and r = 0.478, respectively).ConclusionsPoor blood glucose control for a long time and strengthening inflammatory response are correlated with the status of acute ketosis in type 2 diabetes mellitus patients. The CRP levels in ketosis patients were significantly elevated and could be used to evaluate the degree of ketosis

Wearable sensor-based human activity recognition using hybrid deep learning techniques

Human activity recognition (HAR) can be exploited to great benefits in many applications, including elder care, health care, rehabilitation, entertainment, and monitoring. Many existing techniques, such as deep learning, have been developed for specific activity recognition, but little for the recognition of the transitions between activities. This work proposes a deep learning based scheme that can recognize both specific activities and the transitions between two different activities of short duration and low frequency for health care applications. In this work, we first build a deep convolutional neural network (CNN) for extracting features from the data collected by sensors. Then, the long short-term memory (LTSM) network is used to capture long-term dependencies between two actions to further improve the HAR identification rate. By combing CNN and LSTM, a wearable sensor based model is proposed that can accurately recognize activities and their transitions. The experimental results show that the proposed approach can help improve the recognition rate up to 95.87% and the recognition rate for transitions higher than 80%, which are better than those of most existing similar models over the open HAPT dataset

Epigenetic Profiling of H3K4Me3 Reveals Herbal Medicine Jinfukang-Induced Epigenetic Alteration Is Involved in Anti-Lung Cancer Activity

Traditional Chinese medicine Jinfukang (JFK) has been clinically used for treating lung cancer. To examine whether epigenetic modifications are involved in its anticancer activity, we performed a global profiling analysis of H3K4Me3, an epigenomic marker associated with active gene expression, in JFK-treated lung cancer cells. We identified 11,670 genes with significantly altered status of H3K4Me3 modification following JFK treatment (P<0.05). Gene Ontology analysis indicates that these genes are involved in tumor-related pathways, including pathway in cancer, basal cell carcinoma, apoptosis, induction of programmed cell death, regulation of transcription (DNA-templated), intracellular signal transduction, and regulation of peptidase activity. In particular, we found that the levels of H3K4Me3 at the promoters of SUSD2, CCND2, BCL2A1, and TMEM158 are significantly altered in A549, NCI-H1975, NCI-H1650, and NCI-H2228 cells, when treated with JFK. Collectively, these findings provide the first evidence that the anticancer activity of JFK involves modulation of histone modification at many cancer-related gene loci

Leveraging Nanocavity Harmonics for Control of Optical Processes in 2D Semiconductors

Optical cavities with multiple tunable resonances have the potential to provide unique electromagnetic environments at two or more distinct wavelengths—critical for control of optical processes such as nonlinear generation, entangled photon generation, or photoluminescence (PL) enhancement. Here, we show a plasmonic nanocavity based on a nanopatch antenna design that has two tunable resonant modes in the visible spectrum separated by 350 nm and with line widths of ∼60 nm. The importance of utilizing two resonances simultaneously is demonstrated by integrating monolayer MoS[subscript 2], a two-dimensional semiconductor, into the colloidally synthesized nanocavities. We observe a 2000-fold enhancement in the PL intensity of MoS[subscript 2]—which has intrinsically low absorption and small quantum yield—at room temperature, enabled by the combination of tailored absorption enhancement at the first harmonic and PL quantum-yield enhancement at the fundamental resonance.United States.
